Chemopreventive and antioxidative activity of Carissa carandas extract against chemical induced skin tumorigenesis in mice

Abstract: Objective of the current study was to investigate effect of Carissa carandas extract on skin tumorigenesis, initiated by topical application of 7, 12-Dimethylbenz (a) anthracene (DMBA) and promoted by repeated application croton oil till the end of the experiment ( i.e. 16 weeks) caused a 100% tumor incidence (Group III) in Swiss albino mice. Assess of antioxidants enzymes (SOD and CAT), antioxidative non-enzyme (GSH), vitamin C, Phase II detoxification agent (GST), lipidperoxidation by product and total proteins during DMBA-croton oil induced skin squamous cell carcinoma measured the mechanistic pathway for its chemopreventive effectiveness. Chemo preventive activity of Carissa carandas extract evaluated by decreasing invisible tumour incidence 60% in peri and post initiational phase (i.e. CCE was given 7 days before DMBA application and continued throughout the experiment). Cumulative number of papillomas, tumour burden, tumour yield and average latent period reduced as (23, 3.8, 2.3 and 11.13, respectively) as well as a significant fall in lipid peroxidation and a significant elevation in antioxidant and total protein (p < 0.001) also examined in DMBA/CCE treated experimental mice (Group IV) in comparison to carcinogen treated control. The result demonstrated that administration of CCE have potent suppressing effects on reactive oxygen species during skin carcinogenesis. This is probably due to its modulating effect through inverted the DMBA induced biochemical and morphological changes in the liver and skin both.
