Development Methods for Determination of Bioactive Molecules by HPLC

Abstract: Background and Objective: As regard the cannabinoids, it is known there are two types of cannabinoid receptors defined as CB1 e CB2.The first is expressed principally at central level, while the second is at peripheral level. Recent data indicate that CB2 cannabinoid receptors participate in the control of peripheral pain,osteoporosis,growth of malignant gliomas,tumorsand immunological disorders such as multiple sclerosis. Materials Methods: Examined compounds are heterocycles with a 1,8-naphthyridinic. The work regarded the analytical study of newly synthesized biologically active compounds and, in particular, the development of new methods of analytical separation of structural isomers of new ligands on CB2 cannabinoids receptors. In addition, it was necessary to determine all those parameter indispensable to applying a separation through a preparatory HPLC procedure, eluent mixture, Taking into consideration as the eluent phase a methanol buffer solution, it was determinate, by means of a UV spectrophotometer, the maximum absorption length and therefore the best coefficient of molar extinction of solution of the mixtures under study. In these conditions a wave length of 280 nm was chosen as the absorption maximum in the range far from the visible. Then, I proceeded to determine the best analytic condition, testing various types of silicon Gel C18 columns in inverse phase varying granulometry, length and internal diameter. Results: Isomer cis;Isomer trans;Racemic mixture. Conclusion: In this way each single isomer was obtained with a purity greater than 96%.
