DESIGN DEVELOPMENT AND CHARACTERIZATION OF TRANSDERMAL PATCH OF GRANISETRON HCL.

Abstract: Present research work was aimed to prepare transdermal patch of drug Granisetron HCl with the purpose of developing a dosage form for sustain release of action and improve bioavailability of drug which is beneficial in management of an improve side effect and tolerability profile, lower risk of drug interaction and bypass first pass metabolism. In the present study, transdermal patch of anti-emetic drug was prepared using various polymers, permeation enhancer and plasticizer. Preliminary Selection of type of polymer, preliminary selection of solvent type and volume, preliminary selection of plasticizer type and volume, preliminary selection of formulation, characterization of transdermal patch of drug were carried out. Further, drug patch was evaluated by various parameters. Pre-formulation study of drug and FTIR and DSC study of drug and drug-excipients shows the characteristic peaks and all other peaks observed with individual compound have remained unaffected in formulations indicates Granisetron HCl. Formulation and Development of Granisetron HCl Transdermal Patch Using 32 Factorial Design approach, check point analysis of batches obtained from factorial design and characterization of optimized batch, skin irritation study and stability study of optimized transdermal patch of Granisetron HCl. The Transdermal Patch of Granisetron HCl showed better physical and mechanical properties. It was concluded that Granisetron Patch can be considered suitable for clinical use in the treatment of cancer therapy induced nausea and vomiting.
