Formulation and evaluation of mucoadhesive buccal patch containing captopril

Abstract: Captopril is an antihypertensive drug- angiotensin-converting enzyme (ACE) inhibitor, which exhibits a bioavailability of 50% if taken in the presence of food, this problem can be avoided by the formulating captopril containing buccal patch which may alternatively increase bioavailability by absorbing to the systemic circulation and act on the angiotensin-converting enzyme. In this study, a mucoadhesive buccal patch of captopril was prepared using HPMC K100m, Sodium alginate as mucoadhesive polymer and PEG 400 was used as a plasticizer. The patches were studied for its physical parameter, drug content, mucoadhesive strength, in vitro diffusion, in vitro dissolution and ex vivo permeation studies. The percentage drug content of the formulation (F1-F9) was found to be in the range of 94.5%-98.8%. Fourier transforms infrared spectroscopy (FTIR) and Differential scanning calorimetry (DSC) studies showed that there was no considerable interaction between the drug and polymer used. The release of drugs from all the formulation (F1-F9) in phosphate buffer pH 6.8 was found to be in a sustained manner during an 8hour study. The ex vivo permeation of ramipril from selected formulation (F8) of the buccal patch showed that the drug permeation well across buccal mucosa over a 6hour time period. It may be concluded that the mucoadhesive buccal patch of captopril is an alternative way to bypass hepatic first-pass metabolism or interaction of the drug with food therefore it is expected to improve the bioavailability of captopril.
