HLA - COPD and bronchial asthma markers: commonalities and differences

Abstract: The aim of the work is to study associations of HLA class I - A, B and HLA-DRB1 polymorphic allele genes and their haplotype combinations with chronic obstructive pulmonary disease (COPD) and bronchial asthma in people living in the tobacco growing region of Urgut population of Samarkand region of Uzbekistan. HLA-genotyping was carried out on 49 patients with bronchial asthma, 51 patients with COPD and 90 healthy persons living in the tobacco-growing area of the Uzbek ethnic population. HLA antigens of class I were determined in a standard two-stage micro lymphocytotoxic test by Terasaki (lymphocytes from peripheral blood were isolated by Boyum method). The reaction results were taken into account by the number of deaths of stained lymphocytes. HLA class II genes (DRB1, DQA1, DQB1) were typed by polymerase chain reaction (PCR) according to the manufacturer's method in DNA obtained from peripheral blood nuclear cells (PC) using reagent kits from NPF "DNA-Technology" (Moscow). The degree of association of HLA-alleles and haplotypes with the development of COPD in the tobacco region was determined by the value of relative risk (RR). Negative COPD associations with HLA-DRB1*01 alleles (OR = 0.021; p < 0.001) were detected; HLA-markers of resistance to COPD and bronchial asthma development in tobacco growing region of Uzbek population were determined.
