Synthesis of Prodrugs of Metronidazole to Mask the Bitterness

Abstract: Metronidazole is a known antiprotozoal and antibiotic medication use in the treatment of many serious infections. The bitter taste of metronidazole couldn’t be masked by sugar-coating only, and it needs more effective methods like prodrug, which mask the bitterness through decrease aqueous solubility. Nine mutual prodrugs of metronidazole were synthesized to mask the bitter taste, metronidazole was reacted with different types of non-steroidal anti-inflammatory drugs (NSAIDs) via one of two esterification reactions, either Steglich esterification in the presence of N,N'-Dicyclohexylcarbodiimide(DCC) and 4-Dimethylaminopyridine(DMAP) or after converting the NSAIDs to their corresponding acid chloride in the presence of Thionyl chloride. The synthesized compounds were identified by elemental analysis, infrared spectra (IR), and Proton nuclear magnetic resonance spectra (1H NMR). The purity of synthesized compounds was confirmed by thin layer chromatography (TLC). The aqueous solubility of the synthesized prodrugs was studied, and all the prodrugs show aqueous solubility lower than metronidazole.
