Efficacy of Orthosiphon stamineus aqueous extract against Mercuric Chloride-Induced Nephrotoxicity in Rats

Abstract: Mercurial induced nephrotoxicity is one of the plausible outcomes of the recent progress in the industrial field. It occurs with the inorganic derivatives of mercury due to the tendency of mercurial ion to precipitate in kidneys. This study aimed at finding the protective impact of the alcoholic extract of Orthosiphon stamineus in a model of mercuric chloride induced nephrotoxicity. A group of Wister Kayota rats was fed with food and water contaminated with the non-lethal dose of mercury (0.15 mg HgCl2/kg food or water). Another 3 groups were given different doses of a standardized Orthosiphon stamineus extract at 100. 200 and 400 mg/kg/day P.O (by mouth) every other day for 60 days. Blood samples were withdrawn through out the study to measure the oxidative stress biomarkers, such as; malonyldialdehyde (MDA) , reduced form of glutathione (GSH-Px) and superoxide dismutase (SOD) as well as assessment of the renal function ( serum urea and creatinine). Simultaneous co-administration of the herbal extract (100 -400 mg/kg b.w/ p.o ( by mouth) every other day for 60 days along with mercuric chloride produced a pronounced cardinal nephro-protective effect. In sum, orthosiphone stamineus can be applied as a nephroprotective agent against mercury induced nephrotoxicity.
