Cytotoxic effect of avocado (Persea americana) seed extract containing quercetin in T47D cells by inducing apoptotic pathway in vitro and in silico

Abstract: This research was conducted to determine the cytotoxic activity of avocado (Persea americana) seeds extract containing quercetin in T47D cells by in vitro and evaluate its molecular mechanism that play role in activity as anticancer agent using in silico with Bcl-2, caspase 8 and caspase 9 as target proteins. Cytotoxic activity was carried out using MTT assay method. Extract was given to T47D breast cancer cells with various concentration (10-200 ??g/mL). The absorbance data obtained was converted to average of % cell viability and used to determine IC50. Molecular docking techniques was performed to predict quercetin as active compound in avocado seeds activity against Bcl-2, caspase 8 and caspase 9. This assay step consist of preparation and optimization of chemical compound using hyperchem 8 programs, target proteins preparation using chimera1.11.1, validation process and molecular docking using the AutoDockTools 1.5.6 program. In vitro assay using MTT assay showed higher concentration of avocado seeds extract bring down the viability of T47D cancer cells. Quercetin contained in avocado seeds extract with reflux method has potent cytotoxic activity against T47D cells with IC50 is 54.21 ??g/mL. Quercetin had an activity as a breast anticancer by inhibiting Bcl-2 and inducing caspase 8 and caspase 9. The affinity quercetin with Bcl-2, caspase 8, and caspase 9 is indicated by binding value are -7.12, -5.95 and -6.31 kkal/mol. Avocado seeds extract containing quercetin cytotoxic to T47D cancer cells through increasing the apoptotic mechanism with inhibit Bcl-2 and induce caspase 8 and 9.
