Optimization of Vi Capsular Polysaccharide (Typhoid) Conjugation with Carrier Protein and Characterization of Vi Capsular Polysaccharide Conjugate

Abstract: Development of cost-effective conjugated vaccines is one of the challenges. In the current study their parameters such as pH, Vi polysaccharide to protein ratio and ethylene carbodiimide concentration were optimized. Statistical method such as response surface method (RSM) with Box-Benhan design (BBD) was employed to optimize these parameters. All selected parameters were studied at 3 levels. The lowest % error indicates that the experiments were conducted in a faultless manner. The high regression correlation coefficient (R2) value of 0.9947 indicates that the observed and predicted values has good correlation and the constructed model could be validated. With the help of BBD method of optimization 92% of conjugation was achieved. This is the first report on optimization of conjugation process for typhoid vaccine.
