Synergic antibacterial activity of some wild mushroom extracts with some antibiotics against resistant Gram negative bacteria

Abstract: The rise in drug resistant pathogens has driven the need for combine drug therapy. This study assessed the antimicrobial effect of four mushrooms species in combination with commercial antibiotics against resistant Gram negative bacteria. The crude extracts of mushroom species: Trametes betulina, Pleurotus ostraetus, Agaricus bisporus var. albidus and Agaricus bisporus var. avellaneus, were assessed for their antibacterial effects alone and in combination with antibiotics using disc diffusion method against Pseudomonas aeruginosa, Escherichia coli and Klebsiella pneumonia. The findings showed that crude mushroom extracts alone had antibacterial activity against the test bacteria especially at high concentrations of the extracts and were comparable to some antibiotics. There were synergic effects of the mushroom crude extracts with antibiotics; gentamicin and imipenem on Pseudomonas aeruginosa even at low concentrations while ampicillin only showed synergy at high concentrations of the extracts. Cefotaxime showed the highest zone of inhibition (33mm) on Escherichia coli together with hot water T. betulina and ethanol P. ostreatus extracts. Ampicillin, cefuroxime and gentamicin showed synergy with some of the mushroom extracts on E. coli. T. betulina extract showed additive effect with gentamicin, tetracycline, cefuroxime, chloramphenicol, cefotaxime, imipenem, and ciprofloxacin on Klebsiella pneumonia. The synergistic action was mostly concentration dependent as higher concentration of the mushroom crude extracts showed higher activity in combination with antibiotics. In conclusion, mushrooms extracts were shown to potentiate the activity of some antibiotics against resistant bacteria species and therefore mushroom extract may be an alternative approach in safe drug combination to curb resistant pathogens.
