To study the anticancerous activity of Ajuga bracteosa wall ex. Benth

Abstract: Objective- In now a day’s cancer is also a most common life threatening disease. The major reason is due to uncontrolled growth, differentiation and death of invading cells, organs, tissues. It also occurs due to the drug resistance, toxicity, and low specificity etc. The research work focuses on the development of new herbal drugs to cure the cancer. The present study aims to investigate the cytotoxic potential of Ajuga bracteosa plant extract. In India, the plant is popularly known Neelkanthi, belonging to family Labiatae. Methods- The plant part is converted in to moderately coarse powder form and then was successively extracted with polar solvent like petroleum ether, chloroform, methanol and ethanol solvent and finally the mark were macerated with purified water. These extracts collected and tested against for different types of human cancer like breast adenocarcinoma (MCF-7) and larynx carcinoma (Hep-2) tumor cell lines by using the thiazolyl blue test (MTT) assay. Results- The % yields were found to be 89% by petroleum ether, 15.45% with Methanol, 2.85 % with Ethanol, 15.32 % with Chloroform and 16.18 % with Aqueous extract. In all, the methanolic extract of plant had shown the significant results against MCF-7 and Hep-2 tumor cell lines. The extract of plant like methanolic, petroleum ether and aqueous presented an IC50 value of 10, 65, 70 µg/ml and 5, 30, 15 µg/ml on MCF-7 and Hep-2 cells, respectively at 24 hr. The cytotoxic activity of methanolic extract was found due to the presence of ß-sitosterol. Conclusion- The methanolic extract of plant was found to inhibit the growth in MCF7- breast adenocarcinoma cell line in dose dependent manner with 68.7% and 72.7 % of inhibition in case of Hep-2 larynx carcinoma, respectively as compared to control. The study concluded that ß-sitosterol could have potential benefits for women with a risk for estrogen-dependent breast cancer and thus used for the treatment in the breast adenocarcinoma. Further the research is necessary to study various biological evaluations.
