Caenorhabditis elegans as a model organism for the Evaluation of Anti-hyperlipidemic activity of ethanolic extract of Impatiens Balsamina

Abstract: Objective: The objective of the present study was to evaluate the anti-hyperlipidemic activity of ethanolic extract of Impatiens balsamina using Caenorhabditis elegans as a model organism. Method: The dried seeds of Impatiens balsamina were grinded into fine powder and extracted with ethanol by cold maceration technique for four days. The extract was then subjected to preliminary phytochemical analysis. In-vivo anti-hyperlipidemic activity was evaluated both qualitatively (Lipid staining technique) and quantitatively (Spectrophotometric technique). Caenorhabditis elegans were incubated with normal Cholesterol (5 ppm) and high Cholesterol (10 ppm) containing standard Orlistat (6 µg/mL) and another group containing various concentrations of the plant extract. Anti-hyperlipidemic activity of the extract at different concentrations (10 ppm, 25 ppm, 50 ppm, 100 ppm) was compared to that of standard Olive oil. The absorbance of standard and sample mixtures were measured at 525 nm using in a spectrophotometer. A change in percent of inhibition of lipid accumulation with respect to concentration was compared. Result: Preliminary phytochemical screening showed the presence of alkaloids, flavonoids, tannins, phenol compounds, terpinoids, and saponins. Incubation of the nematodes with higher concentration of Cholesterol (10 ppm) in the medium increased the total cellular lipid, compared to those incubated with standard Cholesterol concentration. Sudan black B staining showed no substantial difference appeared in the fat stores of nematodes incubated with a normal medium. When cultured on a high lipid medium, however, the nematodes were seen to accumulate excess lipid as shown by the appearance of higher number of dark stained lipid droplets. Visually, the lipid droplets in the high lipid control of wild type N2 were more noticeable than the others. Conclusion: The results obtained from the study indicates that the ethanolic extract of Impatiens balsamina possess anti-hyperlipidemic activity which may be due to the presence of flavonoids and other constituents.
