High Performance Liquid Chromatographic Estimation of Doxofylline in Tablet and Syrup Dosage Form

Abstract: A simple, accurate and precise RP-HPLC method was developed for determination of Doxofylline in tablets and syrup dosage form. The chromatographic separation was carried out on a LichroCART LiChrosphere 100; C18 column (250mm × 4mm i.d., 5µm) using methanol: water (50:50, v/v) as mobile phase at flow rate of 0.8ml/min .The UV detection wavelength was 275nm.Xanthinol nicotinate was used as the internal standard. The linearity is obeyed over a concentration range of 0.05-400 µg/ml with a correlation coefficient of 0.9999.The LOD and LOQ were determined for both the tablet and syrup formulations. The proposed method is validated by determining accuracy, precision, stability, specificity and system suitability parameters. The method was used successfully for the determination of Doxofylline in both tablet and syrup dosage form. The advantage of this method is that the mobile phase is simple, method is fast, cost effective and has a wide range of linearity; making it suitable for routine analysis in laboratories.
