Preparation and Physicochemical Characterization of Methotrexate PLGA Microparticles

Abstract: The development of injectable microparticles for anticancer drug delivery is a major challenge. The microparticles were prepared by solvent evaporation method. The physico-chemical characteristics (% yield, drug content, t50) were affected by various parameters e.g. types of disperse phase and continuous phase and excipients of disperse phase was studied. The % practical yield and drug content was significantly influenced by continuous phase. In aqueous continuous phase practical yield and drug content was very low which was improved by substitution of oily continuous phase. From in-vitro release profile % was higher in aqueous continuous phase. It showed biphasic release pattern. The initial burst was due to hydrophilic nature of drug and sustenance in release imparted by lipophilic nature of polymer. This modification of drug release was obtained in appropriate combination of drug polymer ratio which was studied preliminarily.
