Formulation and Evaluation of Tablets of Micronized Glibenclamide (Glyburide) in Industrial Set-Up

Abstract: Up to 3 fold increase in dissolution of glibenclamide from the tablet dosage form could be achieved by employing micronized drug. Milling was done with Cadmill and Fluid Energy Mill (FEM) for different number of cycles. Percentage of fines was found to be highest with FEM-3 cycle. A significant difference in dissolution profile was observed for formulations prepared using different batches of unmicronized and micronized glibenclamide. Formulation F8 showed highest % dissolution (90.27%) in 30 min. as compared to marketed brands X (Daonil-5mg from M/s Aventis Pharma) and Y (Euglucon-5mg from M/s Nicholas) (36.85% and 39.44%). Results indicate that mechanical method of micronization for glibenclamide by Fluid Energy Mill (FEM) was most efficient and cost effective way of achieving particle size reduction and formulation made from it showed
