Anticarcinogenic ,immune modulatory and Antibacterial effect of L-Argininedeiminase ADI against S. epidermidesCoNS mammary gland infection model

Abstract: The study sought to purify a novel L- Arginine deiminase enzyme (ADI) from L.pentosusand studying the effect of L. pentosusand ADI againstmultidrug-resistant S. epidermidesCoNS isolatesin vivoand in vitro. ADI from Lactobacilluspentosuswas extracted,purified andcharacterized.the optimum pH activity was found to be at pH 6.5 ,ADI activity increased when the enzyme was incubated with MnCl2, FeCl2, MgCl2 and HgCl2 with 5 and 10 mM as ADI activitors, which may indicate that this enzyme may exhibit as a metaloenzyme. The molecular weight of purified ADI was 178000 Dalton . The antibacterial activity of purifiedADI was higher than crude (P<0.05). while there isan improvement of S. epidermidesinfection in mammary gland ADI treated groups(P<0.05). Analysis of serum IL-10 and IL-12 levels revealed ADI hasprophylaxis effect. In conclusion, ADI is protein in nature,This enzyme that hydrolyze arginine to generate energy in many microorganisms and it has a potent anticancer activities and can halt growth of solid tumors catalyzes the irreversible hydrolysis of arginine,Lactobacillus pentosusand ADI hasanti-S. epidermides activity, immunomodulatory by increasing pro-inflammatorycytokines and - S. epidermidesin mammary gland infection model in vivoand in vitro.
