Enhanced Analgesic Activity of Diosgenin and Silymarin in Combination on Experimental Models of Nociception in Rats

Abstract: The present study was carried out to evaluate the analgesic activity of Diosgenin (DI) and Silymarin (SY) combination using Eddy’s Hot Plate, Tail immersion and Acetic acid induced writhing tests in albino rats. In hot plate and tail immersion tests, the reaction time was measured at 0 min, 30 min, 60 min and 90 min, compared with control. The independent dose of Diosgenin (100 mg/kg) shown significant (P<0.05 and <0.01) effect only at 90 min, whereas, Silymarin (100 mg/kg) shown significant (P<0.05, <0.01, and <0.001) analgesic effect during the entire test period of 90 min except 0 min. In both the methods, combined dose of Diosgenin and Silymarin (50 mg/kg, each) significantly (P<0.01 and <0.001) increased the reaction time by reducing the pain, equivalent to that of the reference standard, Pentazocine (10 mg/kg). In acetic acid induced writhing test, the test drugs DI (100 mg/kg) and SY (100 mg/kg), significantly (P<0.01) reduced the number of writhes by reducing the abdominal contractions induced by acetic acid when compared with control. Their percentage of inhibition was found to be 31.54 and 38.21, respectively. DI and SY at a dose of 50 mg/kg, each, in combination shown potent analgesic effect (P<0.001) against acetic acid induced writhing with 64% of inhibition. This effect was found to be more or less equivalent to that of the standard drug, Indomethacin (10 mg/kg) which shown 69.79% of inhibition. The results suggested that the combined dose of DI and SY shown enhanced (synergistic) analgesic effect than their independent effect.
