Antibiofilm studies of zerumbone against polymicrobial biofilms of Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Candida albicans.

Abstract: Zerumbone is an essential oil component of Zingiber zerumbet (L.) Smith which has been reported as a potential antimicrobial. Research on exploring its potency as antibiofilm against polymicrobial culture, in vitro has been conducted. Its wide spectrum efficacy on inhibition and eradication towards polymicrobial biofilm and polymicrobial biofilm on catheters of Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, Candida albicans in an aerobic environment has been evaluated. Assays on determining the biofilm inhibition and eradication have been as a microdilution assay. A minimum biofilm inhibition concentration, which caused 50 % inhibition (MBIC50) and a minimum biofilm eradication concentration, which caused 50% eradication (MBEC50) were determined by using crystal violet as staining and read on a microplate reader at 590 nm. Furthermore, an analysis using the Scanning Electron Microscope (SEM) has been conducted to evaluate the mechanism of effect. Results showed that zerumbon 1 % showed antibacterial activity (planktonic) against aerobic microbes: S.aureus, P.aeruginosa, E. coli, and C. albicans were 88.17 ± 0.01, 84.90 ± 0.01, 68.21 ± 0.03, 86.02 ± 0.02, Zerumbone can inhibit and degrade monospecies and polymicrobial biofilm as 24 and 48 h in comparison to control (nystatin and chloramphenicol). SEM analyses confirmed the compound capability to disrupt the extracellular polymeric substance (EPS).
