Evaluation of Biocompatibility of Novel Root Canal Irrigant of Iron Oxide Nanoparticles Functionalized With Glycine and Coated By Chitosan

Abstract: Background: This study aims to evaluate tissue reactions of experimental animals toward iron oxide nanoparticles functionalized with glycine and coated by chitosan (Chi- IONP) implanted in subcutaneous tissue by polyethylene tubes carrier. Methods: Twenty-four albino healthy male rabbits were used in this study. Each rabbit received three polyethylene tubes implanted into dorsal connective tissue and divided into three groups as following: 1- Empty tube (ET) serve as negative control, 2- tube filled with sodium hypochlorite (NaOCl 5.25%) and 3- tube filled with (Chi-IONP). The animals were sacrificed in three periods of time 3rd, 7th and 21 day to collect tissue biopsy specimens after procedure of implantation. Conventional procedure of staining with hematoxylin and eosin (H&E) was done for the specimen. All the specimens were analyzed microscopically and statistically for severity of the inflammatory reaction. Statistical analysis used: Statistical analysis was conducted by Kruskal-Wallis and Mann–Whitney U tests performed by using IBM-SPSS program version 22. Results: All the groups showed sever inflammatory response which reduced over time in different rate. At 3rd day no statistically, significant difference was evident among the groups. At 7th and 21-day intervals, there was high significant difference between NaOCl and the other two groups (Chi- IONP and ET). The ET and Chi-IONP groups showed no significant differences. Conclusion: Chi-IONP solution was well tolerated by the tissue, therefore Chi-IONP solution may be considered biocompatible to living tissues.
