Expression of Quorum Sensing signal abaI gene in Acinetobacter baumannii isolates from clinical and environmental samples in Baghdad hospitals

Abstract: Acinetobacter baumannii is one of the most important causative bacteria of nosocomial infections. Attention turned toward this Gram-negative bacterium due to their extensive resistance to antibiotics. One hundred and fifty samples were collected from different sources from Bahghdad/Iraq hospitals, divided into two groups according to their source: clinical (100) samples and environmental (50) samples, collected from (October 2019) till (March 2020). All of these samples were cultured using specific and differential media including traditional culture media and the specific media CHROMagar Acinetobacter, sixteen (16) isolates of A. baumannii were identified by using microscopic examination, biochemical tests. The identification of (16) isolates confirmed by VITEK-2 system and molecular method using OXA-51 gene which is intrinsic gene belong to this species. The antibiotic sensitivity test recognized for all bacterial isolates and the results showed all of the isolates had Multi-Drug Resistance (MDR). This study showed that 11(68.75%) isolates carrying the quorum sensing gene abaI that matter approved by molecular detection method. Two factors; Streptomycin antibiotic and the time factor was experimented to find their effect on bacterial Q.S as well as to find the effect of Q.S on biofilm formation which is one of the most important bacterial virulence factors, the results showed that there was significant enhancement to biofilm formation for the concentration (1ug) but there was on effect related to the use of (0.1ug) concentration in contrast there was no effect regarding to the streptomycin concentration or regarding to the time factor on the abaI gene expression as well as there was no relationship between the gene expression level and biofilm formation level.
